Structure and sequence of the Drosophila genome
Our laboratory initiated the first large-scale physical mapping of the Drosophila genome
which ultimately culminated in the complete sequence of the genome of D. melanogaster by
Celera, Inc. The initial projects made use of large-fragment clones, particular bacteriophage P1
(PAC) and yeast artificial chromosome (YAC) clones. The most useful for our purposes proved to be
PAC clones containing DNA fragments from D. melanogaster in the size range 75-100 kilobase
pairs. The genomic location from which each of these clones derived was determined by in situ
hybridization with polytene salivary-gland chromsomes. At the completion of the project we had
mapped 2467 PAC and 1193 YAC clones and had achieved virtually complete coverage of the euchromatic
portion of the genome.
